On Identifiability of Nonlinear ODE Models and Applications in Viral Dynamics

چکیده
Ordinary differential equations (ODE) are a powerful tool for modeling dynamic processes with wide applications in a variety of scientific fields. Over the last 2 decades, ODEs have also emerged as a prevailing tool in various biomedical research fields, especially in infectious disease modeling. In practice, it is important and necessary to determine unknown parameters in ODE models based on experimental data. Identifiability analysis is the first step in determing unknown parameters in ODE models and such analysis techniques for nonlinear ODE models are still under development. In this article, we review identifiability analysis methodologies for nonlinear ODE models developed in the past one to two decades, including structural identifiability analysis, practical identifiability analysis and sensitivity-based identifiability analysis. Some advanced topics and ongoing research are also briefly reviewed. Finally, some examples from modeling viral dynamics of HIV, influenza and hepatitis viruses are given to illustrate how to apply these identifiability analysis methods in practice.

An algorithm for finding globally identifiable parameter combinations of nonlinear ODE models using Gröbner Bases.
The parameter identifiability problem for dynamic system ODE models has been extensively studied. Nevertheless, except for linear ODE models, the question of establishing identifiable combinations of parameters when the model is unidentifiable has not received as much attention and the problem is not fully resolved for nonlinear ODEs. Identifiable combinations are useful, for example, for the r...
متن کاملPractical identifiability of HIV dynamics models.
We study the practical identifiability of parameters, i.e., the accuracy of the estimation that can be hoped, in a model of HIV dynamics based on a system of non-linear Ordinary Differential Equations (ODE). This depends on the available information such as the schedule of the measurements, the observed components, and the measurement precision. The number of patients is another way to increase...
متن کاملFeasibility of parameter estimation in hepatitis C viral dynamics models
Methodologies are presented for assessing the feasibility of parameter estimation in nonlinear ordinary di erential equation (ODE) models. These methods are applied to a recent model for hepatitis C viral dynamics. Subset selection is performed on themodel parameters, andmaximum likelihood estimation is conducted using available data from the literature.
متن کاملDefinition of General Operator Space and The s-gap Metric for Measuring Robust Stability of Control Systems with Nonlinear Dynamics
In the recent decades, metrics have been introduced as mathematical tools to determine the robust stability of the closed loop control systems. However, the metrics drawback is their limited applications in the closed loop control systems with nonlinear dynamics. As a solution in the literature, applying the metric theories to the linearized models is suggested. In this paper, we show that usin...
متن کاملHigher-order Lie symmetries in identifiability and predictability analysis of dynamic models.
Parameter estimation in ordinary differential equations (ODEs) has manifold applications not only in physics but also in the life sciences. When estimating the ODE parameters from experimentally observed data, the modeler is frequently concerned with the question of parameter identifiability. The source of parameter nonidentifiability is tightly related to Lie group symmetries. In the present w...
